The increasing resistance to antibiotics in the world is alarming while few new types of antibiotics have been developed in the past 50 years. There is therefore a great need to find new antibacterial substances.
The majority of antibiotics in clinical use work by inhibiting the bacteria’s ability to form a protective cell wall, causing the bacteria to crack (cell lysis). Besides the well-known penicillin, that inhibit enzymes building up the wall, newer antibiotics such as daptomycin or the recently discovered teixobactin bind to a special molecule, lipid II. Lipid II is needed by all bacteria to build up the cell wall. Antibiotics that bind to this cell wall building block are usually very large and complex molecules and therefore more difficult to improve with chemical methods. These molecules are in addition mostly inactive against a group of problematic bacteria, which are surrounded by an additional layer, the outer membrane, that hinders penetration of these antibacterials.
“Lipid II is a very attractive target for new antibiotics. We have identified the first small antibacterial compounds that work by binding to this lipid molecule, and in our study, we found no resistant bacterial mutants, which is very promising,” says Birgitta Henriques Normark, professor at the Department of Microbiology, Tumor and Cell Biology, Karolinska Institutet, and one of the article’s three corresponding authors.
In this study, researchers at Karolinska Institutet and Umeå University in Sweden have tested a large number of chemical compounds for their ability to lyse pneumococci, bacteria that are the most common cause of community-acquired pneumonia. The initial tests were carried out in collaboration with the Chemical Biology Consortium Sweden (CBCS), a national research infrastructure at SciLifeLab. After a careful follow-up of active compounds from this screening, the researchers, in collaboration with the University of Bonn in Germany, found that a group of molecules called THCz inhibits the formation of the cell wall of the bacterium by binding to lipid II. The molecules could also prevent the formation of the sugar capsule that pneumococci need to escape the immune system and to cause disease.
“The advantage of small molecules like these is that they are more easy to change chemically. We hope to be able to change THCz so that the antibacterial effect increases and any negative effects on human cells decrease,” says Fredrik Almqvist, professor at the Department of Chemistry at Umeå University and one of the corresponding authors.
In laboratory experiments, THCz have an antibacterial effect against many antibiotic-resistant bacteria, such as methicillin-resistant staphylococci (MRSA), vancomycin-resistant enterococci (VRE), and penicillin-resistant pneumococci (PNSP). An antibacterial effect was also found against gonococci, which causes gonorrhoea, and mycobacteria, bacteria that can cause severe diseases such as tuberculosis in humans. The researchers were unable to identify any bacteria that developed resistance to THCz in a laboratory environment.
“We will now also initiate attempts to change the THCzmolecule, allowing it to penetrate the outer cell membrane found in some, especially intractable, multi-resistant bacteria,” says Tanja Schneider, professor at the Institute of Pharmaceutical Microbiology at the University of Bonn and one of the corresponding authors.
The research was carried out in close collaboration with Karolinska University Hospital and the University Hospital in Bonn. There were always blockages
In the history of the Federal Republic of Germany, there have been repeated conflict situations between the federal government and the federal states. Especially in the 1970s, when the social-liberal federal governments under Willy Brandt and Helmut Schmidt faced a Union majority in the federal states. And then in the mid-1990s, when the SPD leader at the time, Oskar Lafontaine, overturned the Kohl government’s laws through the state chamber.
The difference to today: While there were clear majorities back then, the picture in the countries today is completely colorful. In the 16 federal states there are at least eleven different color combinations, depending on how you calculate. Black-red, green-black, red-red-green, Jamaica, the traffic light, and a few others. Only one country, Bavaria, does not have any of the three traffic light parties in government.
There is a lot to be said for cooperation instead of confrontation
In this situation, cooperation is the order of the day, not confrontation, according to the political scientist Münch: The federal and state governments, which is already part of German federalism, have to approach each other and make many agreements.

Today is the ninth edition of the North American Leaders Summit in Washington. After a 5-year hiatus, this meeting is finally resumed at the highest level of the leaders of Canada, the United States (US) and Mexico. When this dialogue was created in 2005, it was with the intention of being held annually in a city, geographically rotating the three countries. However, because of the Trump era, this summit was put on hold. Today we could say that the meeting is already late. Why? Because while it is true that Biden assumed the presidency in chaos, it is also true that the priorities have been many, but retaking this summit with his neighbors, friends and partners from the north and south is indispensable. That is why it seems to me that valuable time has been lost this year without having done it before.
There has been a lot of speculation about the issues on the trilateral agenda and although there are official announcements about what will and what will not be discussed, we are certain that one of the great concerns that occupies the governments of Joe Biden and Justin Trudeau is energy reform. presented by the President of Mexico to Congress on September 30. This reform is not only the modification of articles 25, 27 and 28 of the Mexican Constitution, but also represents a direct impact on the value chain of the hydrocarbon sector. In other words, beyond being an issue on the domestic agenda, it is an issue linked to the international commitments established in the TMEC. So no matter how much they say it’s not on the agenda, I say it is.
Another interesting topic is the labor part that has caused so much rash to companies. We will remember that it was an issue full of thorns during the negotiation and that we already knew that although a reform had been made to the Federal Labor Law, we were left lame in this part because there is still much to do.
If we move to climate change, it is not only a multilateral issue where we observed in recent days that the three countries took the roll at COP26 there in Glasgow. I say “roll call” because in many moments, mechanisms and instruments, some countries send officials just to walk, to take their photo and to sign statements. This item is vital because while Canada and the US are aligned with the objectives being achieved, Mexico still has a deficit in the necessary progress. A clear example is the automotive sector. While the TMEC has presented its complexities of interpretation to obtain the content of the regional value in this sector, the supply chain is oriented to conventional automobiles. Another extremely important issue that will be addressed today is that of supply chains. Mexico has expressed its intention to join the supply chains but I have not found any document specifying how. The obvious protagonists of this summit are the three leaders, but there is an invisible protagonist. I am referring to nothing more and nothing less than Xi Jinping, the Chinese leader. There has been a lot of speculation about the issues on the trilateral agenda and although there are official announcements about what will and what will not be discussed, we are certain that one of the great concerns that occupies the governments of Joe Biden and Justin Trudeau is energy reform. presented by the President of Mexico to Congress on September 30. This reform is not only the modification of articles 25, 27 and 28 of the Mexican Constitution, but also represents a direct impact on the value chain of the hydrocarbon sector. In other words, beyond being an issue on the domestic agenda, it is an issue linked to the international commitments established in the TMEC. Another interesting topic is the labor part that has caused so much rash to companies. We will remember that it was an issue full of thorns during the negotiation and that we already knew that although a reform had been made to the Federal Labor Law, we were left lame in this part because there is still much to do.
If we move to climate change, it is not only a multilateral issue where we observed in recent days that the three countries took the roll at COP26 there in Glasgow. I say “roll call” because in many moments, mechanisms and instruments, some countries send officials just to walk, to take their photo and to sign statements. This item is vital because while Canada and the US are aligned with the objectives being achieved, Mexico still has a deficit in the necessary progress. A clear example is the automotive sector. While the TMEC has presented its complexities of interpretation to obtain the content of the regional value in this sector, the supply chain is oriented to conventional automobiles. But these days, the US president said that he would give incentives to not pay the corresponding taxes for the acquisition of electric cars as long as they are made 100% in the American Union. That is, the approach is different.
Another extremely important issue that will be addressed today is that of supply chains. We know that for the tenant of the White House, it is a priority issue and that is why he requested at the beginning of the year an investigation (first of 100 days and then one year) to analyze the products and strategic sectors that are sensitive to dependence on Chinese supplier. Mexico has expressed its intention to join the supply chains but I have not found any document specifying how. Here is a great area of opportunity for our country so that then turn to see how to become the substitute supplier of some products.
The obvious protagonists of this summit are the three leaders, but there is an invisible protagonist. I am referring to nothing more and nothing less than Xi Jinping, the Chinese leader. The bacterial species Escherichia coli (E. coli) constitutes a large fraction of the normal bacterial flora in the intestine. But if E. coli gets into the wrong places in the body, such as the blood or urinary tract, it can cause serious illness. E. coli is the most common cause of urinary tract infections. These can often be treated with antibiotics in tablet form, but some patients become so ill that they must be admitted to hospital and treated with intravenous antibiotics.
“It has long been standard procedure to use the antibiotic cefotaxime for such intravenous treatment. But as time has passed, an increasing fraction of bacteria have become less susceptible to this antibiotic, both in Sweden and the rest of the world, and this has made it necessary to seek an alternative,” says Håkan Hanberger, professor at Linköping University and consultant in infectious diseases at Linköping University Hospital. He has been principal investigator and medically responsible for the study.
The study now published is the result of the Public Health Agency of Sweden being requested by the Swedish government to study how already existing antibiotics can be used in the best way. The researchers have investigated temocillin, a member of the penicillin group of antibiotics that has been known for several decades. It is used in some other European countries, but is not marketed in Sweden. Temocillin acts specifically against E. coli and other intestinal bacteria that can cause urinary tract infections. It is positive that temocillin does not have a broad effect against many different bacteria, since it reduces the risk that the treatment will act against the normal intestinal bacterial flora. This led the researchers to investigate whether temocillin gives less resistance among intestinal bacteria than treatment with the standard antibiotic, cefotaxime.
They studied 152 patients with urinary tract infection that gave rise to fever, also known as pyelonephritis, that required intravenous antibiotics.
“We saw clearly that the intestinal flora was less affected in the group treated with temocillin. The main reason for this is that temocillin gives less selection of resistant intestinal bacteria,” says Charlotta Edlund, professor of microbiology and specialist investigator at the Public Health Agency of Sweden.
The clinical effect of temocillin was as good as the standard treatment with cefotaxime, and the undesired effects were equivalent. The fact that temocillin is less aggressive against the bacterial flora in the intestine suggests that starting to use this forgotten antibiotic for urinary tract infections will have advantages for both patients and society.
“The consequence will be that we see less selection of resistant intestinal bacteria in hospitals, which may contribute to reducing hospital-related infections from these bacteria,” says Håkan Hanberger.

We already know a lot about our enemy, that he can change (mutate), we are equipped with vaccines and most importantly – we are not defenseless. Today, however, fear is not only the virus itself, but also the effects of falling ill with COVID-19. How to protect yourself from pocovid syndrome and make the body fully recover after the disease?
Pocovid syndrome
There are two types pocovid syndrome. The first, the so-called PCS (post COVID-19 syndrome) is defined as the ongoing syndrome of symptoms in patients who have recovered from being infected with COVID-19. The second is Pocovid syndrome in children, the so-called PIMS (pediatric inflammatory multisystem syndrome temporally associated with SARS-CoV-2). As in the first case, it is a complication after COVID-19. The reason is the reaction of the children’s immune system to the presence of the virus. PIMS appears 2-4 weeks after infection and may also be asymptomatic after suffering from the disease.
The causes of pocovid syndrome in adults are:
lung failure
post-infection immunosuppression – that is, decreased immunity,
complications of comorbidities,
pulmonary fibrosis.
Most patients have mild or asymptomatic disease, while 5–8% of infected patients develop respiratory distress syndrome. Does this mean that the vast majority of patients may not consider PCS to be affected? Unfortunately, any survivor can develop pocovid syndrome.
Symptoms of PCS
Let’s take a look at the problems and symptoms that patients with PCS may face:
exercise-induced breathlessness
chronic dry cough,
exercise limitations,
reduced quality of life,
symptoms of heart muscle damage, including heart failure and myocarditis, or the worsening of an existing cardiovascular disease
changes in the sense of smell and taste
the so-called brain fog, described as difficulty concentrating, difficulty concentrating and remembering.
How to deal with pocovid syndrome?
Currently, the role is underlined all over the world physical activity in return to fitness. People who have undergone COVID-19 can benefit from rehabilitation reimbursed by the National Health Fund up to 12 months after the end of COVID-19-related treatment. The program includes, among others, endurance training classes, breathing exercises, effective cough exercises, general fitness exercises, outdoor exercises, terrain therapy, walking training and relaxation training.
The importance of the recovery movement has been proven by British scientists whose research results were published in the journal “Chronic Respiratory Diesease”. The study was completed by 30 participants who took part in rehabilitation classes twice a week for 6 weeks. Everyone has experienced health effects after contracting COVID-19. The program included aerobic exercise, such as walking or treadmill activities, as well as strength training for the arms and legs. The results show a significant improvement in exercise capacity. The subjects felt an improvement in general well-being and cognitive functions. During the program, none of the participants experienced side effects. Therefore, it can be concluded that physical activity individually adjusted to the patient’s health condition is a safe and effective form of rehabilitation in pocovid syndrome.
Another study looked at the effects of COVID-19 on muscle function, exercise, mood and sleep quality. The study included 48 subjects after COVID-19, who were followed up for 12 weeks. Muscle weakness was observed in 35.4% of the participants. The physical activity rate was low in 39.6%, moderate in 33.3%, and high in 27.1% of the participants. Anxiety, depression and poor sleep quality were observed in 33.3%, 29.2% and 50% of the respondents, respectively. This shows how great is the need for intervention and the inclusion of activity and relaxation techniques in convalescents.
Back to activity
There is no single model, based on scientific evidence, that would define the ideal way to get back into activity for every healer. The key to success is caution and gradually increasing the intensity of your training. Individualized exercises based on the patient’s subjective tolerance will allow to obtain the best results and minimize the risk of complications.
In the recommendations for British doctors, we can read recommendations for starting exercises at least seven days after symptoms have resolved, of which a gradual return to pre-disease activity levels or higher is recommended. If the patient was not physically active prior to COVID-19, it is recommended that you make a lifestyle change to become more active. In people who experience recurrence of symptoms such as cough, shortness of breath, palpitations, fever, and changes in the sense of smell and taste, it is recommended to stop exercising, seek medical advice if necessary, and restart the process of returning to activity after the symptoms have resolved. Grading your exercise progression includes lengthening the duration of your training session and increasing the intensity of your exercise.
My advice to convalescents:
set a goal and evaluate your possibilities,
if you have no symptoms, start with walks, light activity and gradually increase your digestion time and training intensity,
measure your strength on intentions – if you experience discomfort and shortness of breath during exercise – consult a doctor and reduce the intensity of training,
were you not active before COVID-19? Now’s a good time to start. At the beginning, walks will be a good solution for you!
You all know how important physical activity is in everyday life. It allows you not only to enjoy a beautiful figure, but also provides endorphins, thanks to which we can stay healthy, influence our immunity or improve efficiency. It is for this reason physical activity is an important element of rehabilitation in the postovid syndrome. Each of you, if you have had COVID-19, may have had different experiences, ranging from no symptoms to hospitalization. Regardless of this, it is recommended for all convalescents to return to activity, based on their individual abilities. Do not postpone health for later!
