The findings, published in the open-access journal The Seismic Record, suggest that earthquakes magnitude 5.0 and larger could cause ground displacement and liquefaction in Salt Lake City that increase the risk of earthquake-related building damage.

As part of the Wasatch Fault Zone, the region has a complex seismic history, with at least 24 large earthquakes occurring in the urbanized parts of the zone over the past 7000 years. Along with previous excavation, borehole and other geophysical studies, the new research also supports the possibility of through-going ruptures across multiple faults in the Wasatch Fault Zone.

Previous excavations in downtown Salt Lake City showed signs of ground deformation, but questions remain about whether “the deformation was related to shaking-induced liquefaction from a more distant earthquake, or was it related to faults that lie beneath downtown,” said Lee Liberty, a geoscientist at Boise State University.

The new study by Liberty and colleagues indicates that the deformation may be the result of active faults beneath the city’s downtown, and that “ground displacement from underlying faults presents a new hazard that should be addressed,” Liberty said.

To learn more about what lies beneath downtown Salt Lake City, the researchers conducted active source experiments along busy urban streets in the area between North Temple and 800 South Streets. During the experiments, a 200-kilogram weight -the seismic source -was dropped at regular intervals to create seismic waves that could be detected by sensors embedded in a fire hose dragged behind the weight.

“We operated during normal business traffic, but we avoided peak traffic times and major commuted roads,” said Liberty. “Our seismic source is pretty powerful, with respect to our offsets and target depth. We relied on off-duty police officers to control traffic and to safely operate.”

Seismic reflection data collected by the experiments helped Liberty and colleagues identify faulted and folded layers of the ancient Lake Bonneville lakebed that were between 13,000 and 30,000 years old. This folded and faulted zone links the East Bench and Warm Springs faults below the city, the researchers concluded.

Further imaging from seismic wave data provided a better picture of the water saturation and sediment properties below the city, bolstering the case that local liquefaction could occur in a large earthquake. In liquefaction, strong shaking reduces the strength of water saturated and loose soils and turns them into a substance more jelly-like than solid.

Previous trenching uncovered evidence of at least one earthquake event downtown in the past 10,000 years. The researchers hope to follow up their study with more work to better define the earthquake history of individual fault strands in Salt Lake City.

Due to the negative repercussion of the fact, Vice President Hamilton Mourão stated in a note that the Navy and the Federal Police would carry out operations in the Madeira River region to discourage the mine.

A SpaceX Falcon 9 rocket carrying NASA’s tiny IXPE X-ray telescope has rolled out to Kennedy Space Center (KSC) Pad 39A for the last time ahead of a planned Thursday, December 9th launch.

Falcon 9 is scheduled to lift off at the start of a 90-minute window that opens at 1am EST (06:00 UTC). The only payload: a first-of-its-kind 330 kg (~730 lb) spacecraft known as the Imaging X-ray Polarimetry Explorer (IXPE) that hopes to analyze the polarization of X-rays to explore black holes, nebulae, and bizarre lighthouse-like dead stars called pulsars in unprecedented detail. The mission is also interesting just for the sheer disparity between the size of the payload and the rocket that will launch it.

As noted, IXPE will weigh about a third of a ton at launch. SpaceX’s Falcon 9, on the other hand, will weigh roughly 550 tons (1.2M lb) when it lifts off, resulting in a truly unusual payload ratio of approximately 1:1700 or 0.06%. However, Falcon 9 will still have to work extremely hard to get IXPE into the correct orbit. That’s because IXPE is designed to operate in an almost exactly equatorial orbit with a zero-degree inclination.

Launching out of Cape Canaveral, which is located 28.5 degrees above the true equator, it’s physically to launch directly into a 0.2-degree equatorial orbit. Instead, a rocket needs to launch into a due-East parking orbit and then perform what’s known as a plane or inclination change once in space. Plane changes are infamous for often being (in terms of rocket performance) one of the most expensive maneuvers one can perform in orbit. That’s certainly the case for IXPE, which will require a 28.5-degree plane change shortly after liftoff.

For Falcon 9, that means that even the tiny ~330 kg IXPE likely still represents about 20-30% of its maximum theoretical performance (1.5-2 tons) for such a mission profile, while the same rocket is otherwise able to launch about 15 tons (33,000 lb) to the same 600 km (373 mi) orbit IXPE is targeting when no plane change is needed. As an example, per a NASA calculator with access to official performance data, Blue Origin says its massive New Glenn rocket – designed to launch more than 40 tons (~90,000 lb) to low Earth orbit (LEO) – can only launch about 2 tons (~4500 lb) to IXPE’s planned orbit

SpaceX is no stranger to launching absurdly small NASA spacecraft, including the ~700 kg (~1500 lb) Double Asteroid Redirection Test (DART) just last month, but IXPE – about 10% lighter than TESS – will be the smallest dedicated payload ever launched by Falcon 9. Following the launch, Falcon 9 booster B1061 will attempt its fifth drone ship landing more than 650 km (400 mi) downrange. Demonstrating just how much more challenging IXPE’s plane change makes an otherwise effortless launch to 600 km, an older and less capable Falcon 9 booster landed just 300 km (185 mi) downrange after launching TESS to an orbit as high as 375,000 km (233,000 mi) – about the same distance between the Earth and Moon.

Between April-May 2020, the researchers conducted telephone interviews with adults across the United States who use cigarettes and/or electronic nicotine delivery systems (ENDS), such as e-cigarettes. Participants in the study were recruited using an campaign on Facebook and Instagram. During this window, nearly 90 percent of the U.S. population experienced some form of state lockdown, with 40 states ordering non-essential businesses to close and 32 states enacting mandatory stay-at-home orders. At the time of their interviews, all participants were voluntarily isolating at home unless required to leave the house.

Nearly all participants reported increased stress related to COVID-19 — namely, fears about the virus, job uncertainty, and the psychological effects of isolation — and described this as the primary driver of increased nicotine and tobacco use. Decreased use, while less common, was prevalent among “social” tobacco users, who cited fewer interpersonal interactions during lockdown and a fear of sharing products.

At the community level, retail access impacted cigarette and ENDS use differently. While cigarettes were universally accessible in essential businesses, such as convenience stores and gas stations, access to preferred ENDS products was more limited, since “vape shops” and other specialty ENDS retailers were typically deemed non-essential and required to close or limit hours. This drove some ENDS users to order their products online, which often resulted in long wait times due to shipping delays, or product backorder as a result of high demand. As a result, some dual users of cigarettes and ENDS increased their use of readily-available cigarettes.

“Pandemic response policies that intentionally or inadvertently restrict access to lower risk products — through availability, supply chains, or even postal service slowdowns — while leaving more harmful products widely accessible may have unintended consequences that should be considered during policy development,” said Daniel Giovenco, PhD, assistant professor of sociomedical sciences at Columbia Mailman School, and the study’s lead author.

Policy considerations

Given that tobacco use behaviors are expected to increase among some individuals during this sustained period of uneasiness, Giovenco and colleagues proposed several key policy recommendations: expansion of cessation resources and services, including their adaptation for remote delivery; establishment and enforcement of smoke-free home rules to protect household members; and enabling equivalent access to lower risk products — such as ENDS and nicotine replacement therapy — to facilitate harm reduction among those who cannot or do not want to quit using nicotine at this time.

“While quantitative, survey-based studies provide valuable insight into changes in tobacco use during lockdown periods, existing research has drawn mixed conclusions. Our approach was the first to qualitatively capture the complex drivers and mechanisms that may help explain varied behavioral shifts,” noted Giovenco. “COVID-19 mitigation strategies to curb transmission will likely continue for the foreseeable future, with many permanently altering elements of the workplace, education, and consumer behaviors. Our findings can help tailor intervention and policy work to address multi-level determinants of tobacco use in the COVID era and the years ahead.”

Between April-May 2020, the researchers conducted telephone interviews with adults across the United States who use cigarettes and/or electronic nicotine delivery systems (ENDS), such as e-cigarettes. Participants in the study were recruited using an campaign on Facebook and Instagram. During this window, nearly 90 percent of the U.S. population experienced some form of state lockdown, with 40 states ordering non-essential businesses to close and 32 states enacting mandatory stay-at-home orders. At the time of their interviews, all participants were voluntarily isolating at home unless required to leave the house.

Nearly all participants reported increased stress related to COVID-19 — namely, fears about the virus, job uncertainty, and the psychological effects of isolation — and described this as the primary driver of increased nicotine and tobacco use. Decreased use, while less common, was prevalent among “social” tobacco users, who cited fewer interpersonal interactions during lockdown and a fear of sharing products.

At the community level, retail access impacted cigarette and ENDS use differently. While cigarettes were universally accessible in essential businesses, such as convenience stores and gas stations, access to preferred ENDS products was more limited, since “vape shops” and other specialty ENDS retailers were typically deemed non-essential and required to close or limit hours. This drove some ENDS users to order their products online, which often resulted in long wait times due to shipping delays, or product backorder as a result of high demand. As a result, some dual users of cigarettes and ENDS increased their use of readily-available cigarettes.

“Pandemic response policies that intentionally or inadvertently restrict access to lower risk products — through availability, supply chains, or even postal service slowdowns — while leaving more harmful products widely accessible may have unintended consequences that should be considered during policy development,” said Daniel Giovenco, PhD, assistant professor of sociomedical sciences at Columbia Mailman School, and the study’s lead author.

Policy considerations

Given that tobacco use behaviors are expected to increase among some individuals during this sustained period of uneasiness, Giovenco and colleagues proposed several key policy recommendations: expansion of cessation resources and services, including their adaptation for remote delivery; establishment and enforcement of smoke-free home rules to protect household members; and enabling equivalent access to lower risk products — such as ENDS and nicotine replacement therapy — to facilitate harm reduction among those who cannot or do not want to quit using nicotine at this time.

“While quantitative, survey-based studies provide valuable insight into changes in tobacco use during lockdown periods, existing research has drawn mixed conclusions. Our approach was the first to qualitatively capture the complex drivers and mechanisms that may help explain varied behavioral shifts,” noted Giovenco. “COVID-19 mitigation strategies to curb transmission will likely continue for the foreseeable future, with many permanently altering elements of the workplace, education, and consumer behaviors. Our findings can help tailor intervention and policy work to address multi-level determinants of tobacco use in the COVID era and the years ahead.”

A key limitation of any of the CRISPR systems, including that from S. aureus, is off-target cleavage of DNA. A guide RNA may bind weakly to a site whose sequence is a close but imperfect match; depending on how close the match is and how tightly the enzyme interacts with the paired gRNA-DNA complex, the enzyme may become activated and cut the DNA wrongly, with potentially harmful consequences.

To explore whether the S. aureus Cas9 could be modified to cleave with higher fidelity to the intended target, the authors generated a range of novel Cas9 mutants and tested their ability to discriminate against imperfect matches while retaining high activity at the intended site. They found one such mutant, which distinguished and rejected single base-pair mismatches between gRNA and DNA, regardless of the target, increasing the fidelity up to 93-fold over the original enzyme. They showed that the mutation affected part of the recognition domain, the region of the enzyme that coordinates contacts between the enzyme and the gRNA-DNA complex. The mutation had the likely effect of weakening those contacts, thus ensuring that only the strongest pairing — which would come from a perfect sequence match — would trigger enzyme activity.

“Avoidance of off-target cleavage is a crucial challenge for development of CRISPR for medical interventions, such as correcting genetic diseases or targeting cancer cells,” Gu said. “Our results point the way to developing potentially safer gene therapy strategies.”
